Bacilli are identified as Gram positive when they have a matrix with multiple layers of peptidoglican forming the cell wall. The violet crystal, the main element of the Gram dye procedure, is retained and stabilised in the matrix, which makes these bacilli gram positive to appear under a violet microscope.
